In conclusion, OPTIMAL is not a new analysis pipeline per se for IMC or any other multi-plexed tissue imaging data, but it does provide a simple framework for assessing any approach. As we convert segmentation data to .FCS file format, it also makes onward analysis accessible.
We also looked at data transformation using different arcsinh cofactor values and used the Fisher Discrimination Ratio, also know as Linear Discriminate Analysis, to determine a value of 1 gave the best statistical separation between low and high parameter values.
