2025 Oncology Highlights:A landmark year for HER2+ breast cancer as T-DXd moves into earlier settings with superior pCR and residual disease outcomes. Meanwhile, NCCN updates have solidified quadruplet therapies in Myeloma and expanded IO combinations in lung and bladder cancers
Will T-DXd + pertuzumab redefine 1L HER2 +mBC treatment? YES.DESTINY-Breast09 (NEJM, Oct 29, 2025):44% reduced risk of progression/deathPFS: 40.7 months (vs 26.9 mo w/ THP) unprecedented >3yrsORR: 85.1%T-DXd+ pertuzumab combo sets a new gold standard. No new safety signals.
Perioperative EV + pembrolizumab may represent a new standard of care for MIBC
Neoadjuvant EV + pembrolizumab followed by RC + PLND and adjuvant EV + pembrolizumab significantly improved EFS, OS, and pCR in cisplatin-ineligible or cisplatin-declining patients with MIBC.
A 400-mg ribociclib starting dose did not match the efficacy of 600 mg in HR+/ERBB2− advanced breast cancer, but had similar progression-free survival and fewer adverse events, supporting 600 mg as the standard dose with reduction to manage toxicity.
Among patients with locoregionally advanced nasopharyngeal carcinoma, toripalimab combination therapy without concurrent cisplatin was a feasible treatment with high efficacy in failure-free survival and low toxicity.

> Could a common allergy med boost #immunotherapy outcomes in advanced #bladdercancer? This study found that antihistamine use alongside atezolizumab was linked to significantly better survival. Exciting, but prospective trials are the next step!
>Those who have criticized....! 😄A retrospective study suggests that adding antihistamines to atezolizumab in 2nd-line advanced urothelial carcinoma may improve overall, cancer-specific, & progression-free survival.! #bladdercancer#immunotherapy >
Neo-N trial: No lead-in nivolumab advantage in TNBC. Study supports shorter chemo regimens, but further trials are needed to compare with standard care. #BreastCancer#MedicalResearch
*“Neo-N trial shows promising pathological complete response rates in triple-negative breast cancer with nivolumab/carboplatin/paclitaxel, though lead-in nivolumab didn't show benefit Supports shorter, non-anthracycline regimens”*. #cancerresearch#immunoth
The recent 12 year follow up of the C9741 trial, shows that even within ER positive disease, those with low endocrine activity(SET2,3) derive benefit from dose dense chemotherapy. Therefore, restricting it solely to TNBC would exclude other patients who could also be benefitted
12-year follow-up of CALBG C9741 trial: Dose-dense chemo shows sustained benefit in node-positive breast cancer vs. conventional. Improved disease-free & overall survival. Low endocrine activity (SET2,3) predicts benefit from dose-dense chemo. #BreastCancer#Oncology#Chemo
Benefits Beyond TNBC:
The CALGB C9741 trial demonstrated that dose-dense chemotherapy offers benefits in both ER-positive and ER-negative breast cancers.
