Yesterday, we presented the results of the LUCAS and CANFIRE trials at #ASH 2024 in San Diego (Poster ID: 199032). Our studies investigated IRAK4 and IL-1β blockade in lower-risk #MDS to address #inflammation-driven ineffective hematopoiesis.

Anne Sophie Kubasch @ASKubasch@UKL_Leipzig presented long-term data showing that MRD-guided pre-emptive treatment with Aza delayed relapse in patients with MRD-positivity in the phase II open-label RELAZA2 study in patients with MDS or AML.
▪️ After median follow-up of 22.5 months (n=95), 63% patients were relapse-free after 6 months (primary endpoint)
▪️ Rate of major MRD response (reachieving MRD negativity)=65%; minor MRD response (absence of major response + no hematological relapse)=35%
▪️ Favourable ELN risk, time from last treatment, and lower MRD level associated with response to Aza
▪️ No new safety signals identified

MRD-guided pre-emptive treatment with Aza was found to be efficacious in preventing/delaying relapse in patients with MRD-positivity:
▪️ 63% patients were relapse-free after 6 months
▪️ Rate of major MRD response (reachieving MRD negativity)=65%; minor MRD response (absence of major response and no hematological relapse)=35%,
However, additional pre-emptive strategies are needed.
