@ManpreetBola@xjwanglab Great question! Unfortunately, this is tricky to answer as it likely depends on the neuromodulator (you have to consider their signaling pathways). Moreover, you also have to consider possible synergistic effects (as has been reported for adenosine A2A receptors and Grp I mGluRs)

@ManpreetBola@xjwanglab Classic solutions with iGluRs (Wang, 1999) get long timescales with recurrence, which thalamus lacks, so synaptic mechanisms are key. Neuromodulators in principle can do something similar, but mGluRs are synapse-specific, so the slow signal is tied to the input that generated it.
@ManpreetBola@xjwanglab The timescale gap is a key feature as opposed to a bug. Because mGluRs are so slow, input from seconds ago influence incoming sensory processing to give you the ability to integrate information across trials which is otherwise hard to obtain.
