Wow! Great work and exciting times! The thought occurred to me that these might be tiny quantum-optical machines via superradiance.

In microtubules, tryptophan networks emit and reabsorb light together in a coordinated burst, almost like a tiny laser pulse. Vaults have a different shape so it may not work the same, but it’s worth checking.

Also, the inside of the vault binds molecules like NAD plus, which cells use to move electrons around. If light energy from the shell reaches that molecule you might have a protein that turns light into chemical work. A simple glow test on whole vaults versus broken apart ones would show if any of this is real.

To push it further, measure steady state quantum yield of intact vaults versus MVP monomers at 280 and 295 nanometer excitation to catch superradiant enhancement. Run femtosecond transient absorption on holo versus apo vaults to see if excitation migrates from the tryptophan lattice to the NAD plus site on picosecond timescales. Model the dipole coupling network from the published phage vault coordinates to find which repeat domain Trps sit at coherent distances. If the dark subradiant states from single excitation theory show up as long lived intermediates, vaults stop looking like cargo containers and start looking like quantum optical machines.
