🔑 Key Findings:
* LIG1 loss is synthetically lethal with PARP inhibition.
* LIG1 is altered in 6% of CRPC patients.
* Combined LIG1 and PARP inhibition triggers replication stress and DNA damage, ultimately leading to apoptosis.
* LIG1-PARP synthetic lethality is conserved across various cancers, including prostate, lung, breast, and colorectal.
* Co-targeting LIG1 and PARP selectively impacts PCa and breast cancer cell viability.
