$LRMR – Q1 data did show a pretty clear dose response. Also bullish is that they seem to have a straightforward regulatory path, with clinical hold removed, inclusion in the new FDA Start program, and anticipated BLA filing at the end of 2025. Some uncertainty over what we will see in 25 mg LOE data at the end of the year. I look forward to seeing them start to dose 50 mg daily, which will be more likely to show good therapeutic effect sizes. Stock still seems cheap IMO.
$INSM – Q2 Brenso results were excellent, going beyond exacerbation reduction to functional improvements at the high dose. Valuation now seems a bit stretched, and might need to retrace a bit, but excited to see this company mature into a robust biotech with three late-stage / commercial programs with good economic prospects.
$PYXS – CEO revealed that data probably wasn’t enough for POC, and so they extended the timeline to data release to 2nd half. This adds risk, because it’s less likely the early results are exceptional. They are hinting at responses, however, and there are other reasons to believe the drug might be active. Even without eye-popping ORR numbers, if there is some activity, and the tolerability profile is good, it might be “good enough”, because as a combo agent to debulk high stroma tissues with another ADC or an IO agent this could have significant scarcity value. Still cautiously optimistic on this readout in 2H but risky.
$LIFE (now $ATYR) – Instituting the EAP and the number of blinded patients coming fully of steroids are bullish signs for their P3 program. Plus they have added more supportive mechanistic preclinical data. Stock still seems very undervalued. P3 enrollment complete any day and P2 scleroderma data at the end of the year so stock could start to garner attention in 2H.
This new review paper from Chris Voigt's lab is a must-read for #synbio researchers and investors. His keynote at #SEED2024 contained a lot of the case studies and thinking in this paper.

$NWBO Here are my notes on Linda Power’s Q&A session of the ASM. Due to the amount of information I’ll be breaking this up in 2 separate posts. This post is what the company has been working on for the past 18 months. Reminder I am not a legal, financial or medical advisor.
The progress of cancer genetics
A Nature piece argues for classifying cancers by their molecular features (eg. TP53 mutation, PDL1 expression) instead of their organ of origin (eg. lung, breast), because such molecular features are what largely determine the course of treatment.
This reminds me of pure computational biology methodology happening first around ~2010s of clustering patient samples while disregarding their tissue of origin, with the idea of "finding drugs from some cancer types that can be used in different cancer types with fewer treatment options". The input data there was either bulk RNASeq or DNA whole exome sequencing.
Such work would (almost) never make it to top journals at that time, because it was thought that putting together cancers of different types made no medical sense and was simply a theoretical exercise.
Well, that theoretical exercise was the beginning of cancer genomics, which is now the main pillar driving both cancer therapeutics and cancer research forward. So, it wasn't purely theoretical after all.
As this Nature article outlines, classifying tumors by molecular features makes sense from many perspectives, with the most important being getting patients faster access to new drugs with less bureaucratic delay triggered by new clinical trials. Why is this? Because the success of a cancer drug in a patient population is intrinsically centered around the molecular characteristics of tumors. For a new therapy, some patients will respond, others will not. Why is that? The answer (or at least a part of it) is oftentimes hidden in the genome/transcriptome/epigenome of the tumor.
There is however lots of specific (and practical) knowledge being accumulated in the field by people studying a single cancer type in great depth. Oftentimes metastatic cancers originating from the same organ retain similar characteristics to the cells they disseminated from. Therefore completely forgetting about their tissue of origin makes equally little sense.

Good news re the #BIOSECURE act, the House just published new text that now includes a grandfathering for existing contracts/products until Jan 1, 2032, giving the industry ~8 years to switch manufacturing partners and services. There were huge concerns that this process would be rushed. @IAmBiotech did great work on this!
