I am excited to share our work in which we conducted a CRISPRi-based screen to assess 102 Autism Spectrum Disorder (ASD) risk genes in microglial uptake of synaptic material! (1/7)

CRISPR has the potential to cure many genetic diseases, but it’s notoriously hard to use in neurons.
In this work we tackled 2 of the main hurdles: Cas9 delivery and DNA repair.
And we discovered…
The rules of genome editing are different in neurons👇🏾

Microglia activation is commonly seen in ASD and abnormal microglial synaptic remodeling has been speculated to contribute to ASD biology. While expression of ASD risk genes is enriched in neurons, we were surprised to see that microglia express many ASD risk genes! (2/7)
We dove deeply into characterizing how ADNP affects microglia. We found loss of ADNP leads to decreased endocytosis, a remodeled surface proteome, and increased motility. (6/7)
UCSF's Institute for Neurodegenerative Diseases is hosting an open house on November 15, 4-6pm for interested rotation students. Come hear flash talks from each lab and afterward socialize with lab members! See you there!

Yes! In addition to upholding transparency and truly promoting scientific dialogue, this new publishing landscape will stand as a great training resource - reading peer reviews will surely help trainees, like myself, think critically about their own research
Today, we’re introducing a new model that eliminates accept/reject decisions.
By publishing every paper with eLife reviews as a Reviewed Preprint, we plan to restore autonomy to authors, ensuring that they will be judged by what, not where, they publish. https://t.co/OAsiOVFStI
Are you a future postdoc or incoming @UCSF grad student interested in neurodegenerative disease?
Join the Open House of the @INDUCSF to learn about our research and meet PIs and current trainees.
