#ASCO25 ctDNA from the CALLA Study of cervical cancer take aways:
1) baseline detection of ctDNA for pts with local regionally advanced disease is 99%
2) Among pts on the durva arm, at cycle 3, 38% of pts had "+" ctDNA and by cycle 6 this dropped to 23%. (figure 1)
3) pts with "-" ctDNA looked like they did NOT benefit from durva but in the ctDNA "+" there is a signal of benefit (figure 2 and 3)
4) ctDNA "+" at cycle 3 was associated with recurrence in 68% of patients with a median lead time of 164 days (figure 4)
My question: If I know you are ctDNA+ at cycle 3 (?or 6 6) should we add additional therapy aka OUTBACK or a new ADC at that point? or wait until clinical progression? What changes OS? What changes patient QoL?

⏯️ Check out this new video showing the ability of NeXT Personal® to consistently achieve highly sensitive limits of detection (LOD) across a large number of patient samples across many solid tumor types, including samples of poor quality:
• NeXT Personal demonstrated a median LOD of 1.9 parts per million (PPM) with a range of 1.6-2.8 PPM across this real world data cohort (4,232 plasma timepoints across 1,796 patients)
• 40.9% of all MRD positive samples were detected in the ultrasensitive range (≤100 PPM), and 86.8% of molecular recurrence detections were in the ultrasensitive range
• Median ctDNA level for first molecular recurrence detection was 12 PPM, underscoring the importance of ultrasensitive testing for early recurrence detection

Transforming bladder cancer immunotherapy-monitoring treatment decisions through molecular insights: Dr. Alan Tan, MD's (@alantanmd) recent presentation showcases how ctDNA analysis with our @TempusAI /Personalis xM (NeXT Personal® Dx) assay provided predictive information for his patients.
Patient Vignette 1: ctDNA Kinetics were Predictive
• Patient with muscle-invasive bladder cancer showed initial ctDNA decrease during neoadjuvant therapy (from ~1000 to 23 parts per million)
• Critical finding: ctDNA did not clear on IO (immunotherapy) neoadjuvant treatment with enfortumab vedotin and pembrolizumab (EV/P), with the low level of ctDNA detection in the ultrasensitive range. The patient then showed an upward trend in ctDNA pre-cystectomy.
• Surgical outcome: Surgery confirmed lymph node involvement and ovarian spread
Our groundbreaking data in #BreastCancer was just published in the ESMO Annals of Oncology! (@myESMO).
In collaboration with @royalmarsdenNHS and @ICR_London (@igmurillas), this study reports on the performance of NeXT Personal® in a cohort of 78 patients with breast cancer (all subtypes):
- NeXT Personal detected very small traces of cancer recurrence in the blood a median of ~15 months ahead of scans
- The test detected 100% of the patients that had breast cancer recurrence
- 100% of patients with undetectable post-treatment ctDNA remained recurrence-free during a median follow up period of 76 months.
- NeXT Personal showed high sensitivity for detecting early-stage breast cancer with ~39% of all detections occurring in the ultra-sensitive range below 100 PPM

In collaboration with @CharlesSwanton and his colleagues at @TheCrick and @uclcancer, this groundbreaking study highlights advancements in #LungCancer detection using NeXT Personal®.
Key highlights:
- NeXT Personal test showed high sensitivity for detecting early-stage I-III NSCLC pre-operatively: 100% of non-adenocarcinomas and 81% of lung adenocarcinomas (LUAD)
- Early-stage LUAD patients who tested negative for #ctDNA with NeXT Personal prior to surgery exhibited a 100% 5-year overall survival rate; patients testing positive had a high overall risk of relapse during that same period
- Patients who tested positive for very low traces of cancer (<80 PPM) still had a high risk of recurrence, suggesting the importance of ultra-sensitive #MRDTesting with NeXT Personal
