What's your favorite quick-and-dirty lab technique that still works for you?

I make E. coli electrocompetent cells straight from plate streakouts (no pre-culture in liquid) 😱 at room temperature 😱 with just water (no glycerol) 😱. Success rate >90%, don't @ me
