Women are four times more likely to develop autoimmune diseases, linked to having two X chromosomes.
These findings will enable better screening and earlier detection of autoimmune risk in women.
Background
1/ Autoimmune diseases affect up to 50 million Americans, 4 out of each 5 people with an autoimmune disease are women.
2/ Women have 2 X chromosomes, one is randomly silenced for maintaining the right โdosageโ of X-chromosome gene expression, this is called X-chromosome inactivation.
3/ X-chromosome inactivation relies on a long non-coding RNA called Xist lncRNA.
4/ Xist lncRNA is expressed only in females (males have only one X chromosome, which is active)
5/ Xist lncRNA forms a complex with up to 81 proteins, called Xist Ribonucleoproteins (Xist RNP).
Findings
5/ A 2024 study in mice found that Xist RNP expression in male mice increased their susceptibility to autoimmunity on pars with female levels, including autoimmune multi-organ failure and induced lupus.
6/ It was also found that the reaction to Xist RNP across the body reprogrammed immune T and B cell populations in females, similar reprogramming was seen in transgenic male mice expressing Xist RNP.
7/ Autoantibodies against Xist RNP were also identified in female autoimmunity patients, showing that a similar Xist RNP mechanism underlies female susceptibility to autoimmunity in humans as well.
8/ This is in line with earlier findings showing that T cells in female systemic lupus patients had dysregulated Xist RNP expression.
Significance
9/ The results highlight a new path for diagnosing autoimmune diseases and identifying females at increased risk of developing autoimmunity based on the presence of anti-Xist RNPs autoantibodies.
