Prempeh College was established in February 1949 with 50 students transferred from Mfantsipim School in Cape Coast, Ghana, along with their headmaster, Rev. Sidney N. Pearson. The school opened on premises donated by Asantehene Otumfuo Osei Agyeman Prempeh II and was initially housed at the site of the 52nd Military Hospital in Kumasi.
Its founding was a collaborative effort between the British Colonial Government, the Methodist and Presbyterian churches, and the Asantehene to address the lack of secondary education options in the Ashanti and Northern regions. Prior to this, students from those areas had to travel south to schools like Mfantsipim, Achimota, or Adisadel.

According to reports, Kusi Ernest's friend allegedly poisoned him over a dispute regarding lotto numbers they had staked. The deceased won GH¢100,000, while his friend won only GH¢1,800.
The suspect, Kusi Ernest’s friend, allegedly blamed him for not convincing him to stake more money on the winning numbers and subsequently poisoned him when they went to celebrate their wins at a bar.
