Using a CRISPR screen and single cell analyses we asked how do stem cell-derived islets (SC-islets) react to immunological pressure (allogeneic). Among the hits we identified CXCL10 depletion as a way to escape immune rejection of grafted #betacells.

Couldn’t ask for a better closure for my postdoc era. Was so great to attend and present at #ISSCR2022! Met so many old and new friends/colleagues. Some of which you see only in meetings like these every few years. Hope to see you all next year in Boston #ISSCR2023@ISSCR
Tomorrow 6/16 at #ISSCR2022, excited to present 'Immunogenicity of stem-cell derived islets as defined be a whole-genome CRISPR screen'. In person or virtual.
1:45pm PDT/4:45pm EDT, at 'Stem Cell Bioengineering - enhancing cell function'.
Happy to share a pre-print of some work from my time in the @TheMeltonLab. First and foremost, many thanks to all the co-authors involved!! Now, for a brief outline of the findings..
Glad and excited to share our online pre-print of a paper submitted for peer review in @CellStemCell. We report a whole genome CRISPR screen and transcriptomic profiling of immune challenged stem cell derived islet cells. See thread below

🔬Today in @stemcellreports, research from @naleite, Douglas Melton & colleagues has identified new ways to enhance the SC-beta cell survivability through genetic manipulation of the cells. Read on 👉 https://t.co/SU704vx0KR. Image: The Foundry. @martinperaJAX@TheMeltonLab
“The promising results we’ve seen with #stemcell-derived islet cells could deliver a life-changing therapy for people who suffer from the relentless life-long burden of #Type1diabetes," said Dr. Douglas Melton, co-director of @harvardstemcell Institute.
Are your cells mature? Does it matter? In our latest @CellCellPress, we review Cell maturation: Hallmarks, triggers, and manipulation. We discuss a new framework, based on cell plasticity, to understand how cells become specialized.. @PennCDB@IRM_UPenn
