Report @CCR_AACR on phase II study of cabozantinib in ROS1 NSCLC showed RR 43% including deep response in pt with ROS1 D2033N mutation. Cabozantinib is a type II ROS1 inhibitor and may have a role for pts with on-target resistance to a type I inhibitor.

RASolute 309 Begins in First-Line RAS G12D Metastatic PDAC

On October 5, 2026, Revolution Medicines announced that it had begun treating patients in RASolute 309 (NCT07805954), a global, randomized, open-label Phase 3 trial evaluating RASONQUE (daraxonrasib) plus zoldonrasib as first-line treatment for adults with metastatic RAS G12D pancreatic adenocarcinoma.

The study is comparing RASONQUE plus zoldonrasib with gemcitabine plus nab-paclitaxel. The dual primary endpoints are progression-free survival and overall survival, with key secondary endpoints including objective response rate, duration of response, safety and tolerability, pharmacokinetics, and patient-reported outcomes.

According to Alan Sandler, MD, Chief Development Officer of Revolution Medicines, RASolute 309 is the first Phase 3 study evaluating a RAS(ON) inhibitor doublet approach.
