The MOIO randomised phase 3 non-inferiority trial (n=340) explored early discontinuation of ICI therapy be SOC in advanced patients responding to therapy. It had many flaws (mixed tumors and treatments, small size, early discontinuation etc). Also, due to the statistics, it’s not possible to say whether non-inferiority was achieved (OS HR 1.89 [95%Cl: 1.03,3.48]). Despite these problems it highlights a concern around early discontinuation, which should be taken seriously. Tumour specific trials are needed, but I’m not sure it will be easy to randomize patients . #ESMO26 @OncoAlert@myESMO@Annals_Oncology
New study results from @Cecchini_OncMD show Telisotuzumab adizutecan + bevacizumab appeared to improve efficacy outcomes with a manageable safety profile in patients with advanced #colorectalcancer, and the regimen is currently being evaluated in a Phase III trial, AndroMETa-560. #ESMO26

Zecdarasib (G12Di) plus chemo in 1L metastatic PDAC, phase 1
ORR 89% with mFOLFIRINOX (n=27) ORR 69% with gem/nab-paclitaxel (n=26)
Awesome safety for early DE.
For context, other early 1L PDAC response rates (cross-trial which we would neeeeever do):
Zoldonrasib + FOLFIRINOX: 82% Zoldonrasib + gem/nab-P: 61% Daraxonrasib + gem/nab-P: 59% Daraxonrasib alone: 47%
Chemo clearly still matters. Starting to think about this like Breakwater…
In PDAC, monoallelic inhibitors are extremely effective, just more effective with a little extra oomph behind them.
They will land in adjuvant, neoadjuvant, metastatic and every space you can imagine in the next few years.
Question is do we combo single plus pan or single plus chemo, and what is the added agent (especially in CRC)?

🚀 Presidential session #3
📌 MOIO is an academic, multicentre, phase III non-inferiority trial conducted across 36 French centres.
🗣️ Dr. Gravis
👫🏽 Pts with locally advanced/metastatic solid tumors in partial or complete response (CR) after 6 months of standard-dose IO were eligible (melanoma in CR excluded)
👉🏽 14 different tumor types
👉🏽 Multiple #ICI combinations
👉🏽👉🏽 Inconclusive study!
❓ Worse OS for NSLC and urothelial
❓ Better OS in HNSCC and RCC
⚠️Are we ok for a HR of 1.3 for non-inferiority threshold?

Dr. Gil Morgan (@OncoAlert) breaks down the 6-year NATALEE update!
In this episode of #OAOnTheGO, Dr. Gil Morgan, OncoAlert representative, breaks down the 6-year NATALEE update, simultaneously published in Annals of Oncology.
🔬 Key takeaways:
📊 Overall survival: HR 0.81, with a 2.1% absolute benefit at 6 years.
📊 Invasive disease-free survival: Absolute benefit has widened to 5.3%.
📊 No new safety signals, with patients now more than 3 years off ribociclib.
These findings strengthen the role of adjuvant ribociclib for eligible patients with high-risk, HR-positive, HER2-negative early breast cancer.
While the overall survival analysis remains descriptive and the absolute benefit is modest, the results bring an important question into focus: Can preventing more recurrences ultimately help more patients live longer?
