Very pleased to share our work on development of natural language processing pipeline to assess cardiovascular risk and thrombotic complications in MPN. Machine learning approach allowing assessment of health records of >900 patients and >23000 documents

Proud to share our recent study published on Leukaemia.
QRISK3 score can identify MPN patients with higher risk of TE based on CV risk factors that may benefit of early cytoreduction

Analysis highlighted increased thrombotic risk in PV patients compared with ET and identified hypertension as comorbidity of particular significance for VTE and CVA. Excellent work from @ADuminuco and Josh Au Yeung on getting this project completed.
- STAT5 inhibition by dasatinib dependent on reduction in IL-2
- TIM3+ cells show increase TCR signalling in pts on other TKI 2/3
Thanks to support from @dr_de_Lavallade@DillonHaem @DeeptiRadia @DrLornan@SKordasti@harrisoncn1 and all participating patients and families 3/3
Out now in @Haematologica our paper on effects of TKIs on T cell signalling in patients with chronic myeloid leukaemia.
https://t.co/buDqCbjZ3Q
Dasatinib inhibits TCR and STAT5 signalling and inhibits T cell effector function cf. other TKIs. 1/3
- immunostimulatory effects of dasatinib potentially explained by preferential inhibition of Treg TCR signalling cf effector cells
- pts with clonal CD8+ lymphocytosis also show greater inhibition of Treg signalling, may explain improved leukaemia related outcomes in this group

Finally in print @BloodAdvances, data showing importance of third dose of vaccine for Omicron variant neutralisation as well as increased T cell response against Covid-19 in patients with #MPNsm and #CMLsm
Blood transfusion clinical fellow required to join the team @KHPHaematology @GSTTnhs @KingsCollegeNHS to look at data driven transfusion practice in partnership with the @DataDrivenBTRU team https://t.co/JaFkeEis8o
Increased immune checkpoint expression also seen in patients with refractory disease. Significant increase in plasma pro-inflammatory cytokines at diagnosis. Likely due to LAA overload with increased disease burden. Potential future role for checkpoint inhibitor therapy? 2/2
Paper out in BJHaem @BritSocHaem@G_ICEMD Patients with CML at diagnosis have increased frequency of effector Tregs and Tregs as well as increased expression of T cell exhaustion markers including TIM-3 and LAG-3 https://t.co/Hb43VEwDCU
Pleased to share our research recently published in @BloodCancerJnl highlighting impaired cellular and humoral response to SARS-CoV-2 vaccination in patients treated with ruxolitinib amongst cohort of MPN/CML pts

Other key findings include reduced response in patients receiving ChAdOx-1 vaccine after 2 doses. Presence of comorbidities also associated with reduced antibody response on multivariate analysis.
A 2nd report today from @ljiresearch also demonstrates vaccination induced T-cells vs Omicron (B.1.1,519) well preserved compared with all variants and with 3 different vaccines
