Part of my postdoctoral work in the Ronald lab is on bioRxiv! We applied directed evolution to engineer a new-to-nature immune recognition range in a rice immune receptor
We evolved immune receptor binding to two evolutionarily divergent groups of ligands from a fungal pathogen. Such a receptor would protect from a broader spectrum of pathogen strains and reduce the likelihood that the engineered receptor will be "overcome" by pathogen evolution
This shows how directed evolution can explore large synthetic sequence spaces to develop immune recognition of divergent pathogen ligands, a function that seemed initially far-fetched
-TnpBย (400aa small Cas12-like nuclease) can be used to genome edit plants
-Viral delivery of TnpB+guide using tobacco rattle virus led to somatic edits in arabidopsis
-Efficiency varied by plant and gRNA but up to 80% and there was some germline transmission
