Researchers have uncovered a previously unknown immune structure located within the human skull.
A team of biologists from Washington University School of Medicine in St. Louis has located an overlooked immune organ embedded in the bone marrow of the cranium. For many years scientists assumed that the brain’s protective immune mechanisms operated in complete isolation from the rest of the body, thereby shielding delicate neural tissue from potentially harmful immune activity.
The new findings demonstrate that minute vascular passages extend from the brain, cross the outermost protective membrane, and connect directly with the marrow inside the skull.
Using these pathways, the investigators followed molecular signals into formations that closely resemble the germinal centers of lymph nodes. These sites function as specialized training zones in which immune cells proliferate, identify targets, and prepare to combat invaders at the earliest stage of an assault.
To examine the practical importance of the system, the scientists altered the lymphoid formations in mice bearing glioma, an especially aggressive form of brain cancer. When they stimulated the skull-based immune centers by applying a hydrogel containing specific proteins beneath the scalp, the animals mounted a markedly stronger anti-tumor response and lived significantly longer. In contrast, interference with the same structures allowed the tumors to expand without restraint.
Because comparable follicular helper T cells have already been observed in human skull marrow, the investigators believe analogous defensive hubs are present in people. Should further work confirm this, the discovery may transform treatment approaches for brain tumors, Alzheimer’s disease, and Parkinson’s disease by enabling physicians to stimulate cranial immune centers non-invasively through the scalp and thereby avoid invasive brain operations.
