DDW2026の振り返りで、IBD・SAP関連見つけました。
かなり奥深い研究なので投稿します。

発表番号 Su1515。

対象は潰瘍性大腸炎。自己組織化抗炎症性ペプチド両親媒性分子、AIF-PAsを、IL-2欠損マウスの自然発症性大腸炎モデルに局所投与しています。
SAP‐抗炎症ペプチド複合体です。
投与は結腸洗浄法。
5週齢と10週齢で投与し、15週齢で評価。
つまり、腸管局所に自己組織化ペプチド材料を届け、炎症と粘膜障害を制御できるかを見た研究です。
結果はかなり興味深いです。
AIF-PA1 / AIF-PA2投与群では、腸管の陰窩構造が回復。
対照群では炎症細胞浸潤、腺構造の乱れ、杯細胞の減少が見られましたが、AIF-PA群では組織構造の改善が示されています。これは単なる抗炎症ではなく、粘膜・組織修復の方向です。
機能面でも改善しています。
大腸通過時間は、
生理食塩水群：210.8 ± 9.1秒
対照AIFC-PA6群：248 ± 28.5秒
に対して、
AIF-PA1群：147.5 ± 7.6秒
AIF-PA2群：100.0 ± 13.1秒 有意に短縮。
ペプチド材料によって、生理機能の回復が示唆されています。
免疫環境も変わっています。
CD11c陽性樹状細胞浸潤は、
生理食塩水群：11.8 ± 0.4%
対照AIFC-PA6群：10.4 ± 2.3%
AIF-PA2群：7.5 ± 1.9%
AIF-PA1群：1.4 ± 0.2%
特にAIF-PA1で大きく低下。
一方、抗炎症性のIL-10陽性細胞は増加。
AIF-PA1群：3.6 ± 0.3%
AIF-PA2群：1.9 ± 0.4%
対照群は0.7〜0.8%程度。
つまり、この発表のメッセージは明確です。
自己組織化ペプチド材料が、
・腸管組織構造を改善
・炎症細胞浸潤を低下
・IL-10陽性細胞を増加
・大腸機能を改善 する可能性を示した。
IBDにおいて、自己組織化ペプチドは単なるDDSではなく、局所炎症環境を変える“治療材料”として見られ始めています。
これは突然出てきた話ではありません。
背景には、2021年のAdvanced Therapeutics論文があります。

このSAP-抗炎症ペプチドの研究は、最近の総説でも取り上げられています。
Nature系の npj Biomedical Innovations のレビューでは、粘膜部位の慢性炎症に対する免疫調節バイオマテリアルとして、Sharma / Stuppグループの自己組織化ペプチドナノファイバーが紹介されています。
IBDは、薬剤だけでなく、粘膜免疫・局所炎症・組織修復を制御する“材料科学”のテーマになりつつあります。

そして、もう一つ大きな流れがあります。
これがすごいんです。本当にすごいんです。
これまでも中国系のSAP論文には圧倒されてきましたが、中国科学院系のXuehai Yan教授らが、Advanced Functional Materialsに大型レビューを発表していました。見過ごしていました。ジャーナルインパクトファクターも当然すごいんです。

自己組織化ペプチド、ペプチド共集合体、coacervate、peptide glass、AI-assisted peptide designまで含め、炎症性疾患に対するペプチド材料を体系化しています。

これは中国トップ中のトップが、この領域を正面から取りに来ているわけです。アカデミア世界No1の中国ですよ。
ただ、私はこれを3Dマトリックスにとって単純な脅威とは見ていません。
むしろ逆です。自己組織化ペプチドが、止血だけでなく、炎症制御・粘膜保護・DDS・組織修復へ広がるという世界的潮流が、ここまで明確になってきたわけです。
これは、3Dマトリックスが持つRADA16/PuraStatの素材価値を、外部のトップサイエンスが裏打ちし始めたということです。
もちろん、Sharma / Stupp系のAIF-PAと、3DマトリックスのRADA16/PuraStatは同じものではありません。
しかし共通する本質があります。
それは、自己組織化ペプチドを使って、局所に留まり、炎症環境を変え、粘膜・組織修復を助けることです。
3Dマトリックスにとって重要なのは、PuraStatを“止血材”だけで終わらせないことですね。
