#Lepu Biopharma (as the licensor) and #Excalipoint (as the licensee) have reached a #licensing agreement for two preclinical assets, #CTM012 and #CTM013, developed under Lepu’s proprietary T-cell engager TOPAbody platform.
CTM012 and CTM013 both are #TCE, target undisclosed. CTM012 submitted IND in #China
Under the agreement, Excalipoint will obtain exclusive global rights to develop and commercialize the target assets.
Lepu Biopharma will receive an upfront cash payment of $10 million and ordinary shares issued by Excalipoint Cayman to Innocube, a wholly-owned subsidiary of Lepu Biopharma, representing 10% of Excalipoint Cayman’s expanded issued share capital.
Additionally, Lepu Biopharma is entitled to up to $847.5 million in development and commercial milestone payments, as well as tiered sales royalties ranging from low to mid-single-digit percentages.
Concurrently, Excalipoint Cayman will conduct a Series A financing, with investors including Sequoia China, YuanBio Venture Capital, and Apricot Capital collectively investing $41 million in Excalipoint Cayman.
This transaction will advance Lepu Biopharma’s strategic research and development in next-generation ADC drugs and immuno-oncology bi/trispecific antibodies.
🇨🇳 #Lepu Biopharma recently registered a Phase 3 clinical trial (CTR20253026) on 🇨🇳 #CDE for its TF #ADC #MRG004A, combined with best supportive care (BSC), compared to placebo plus BSC, in patients with advanced pancreatic cancer.
The inclusion criteria is a confirmed diagnosis of unresectable or metastatic pancreatic cancer, with patients having previously received systemic chemotherapy regimens containing gemcitabine and fluoropyrimidines.
The primary clinical endpoint is overall survival (OS), with a planned enrollment of 231 patients.
MRG004A is an ADC drug targeting tissue factor (TF) , linking a TF-targeting monoclonal antibody to the highly potent microtubule inhibitor MMAE via Glyco Connect™ site-specific conjugation and Hydra Space™ polar spacer technology.
It is the first TF ADC drug to enter clinical trials in China.
During the 2024 ASCO, MRG004A’s Phase 1 data were presented.
In the 2.0 mg/kg dose group, among 12 evaluable pancreatic cancer patients (with a median of three prior lines of therapy), 4 PR and 6 SD. ORR was 33.3% (4/12), and DCR was 83.3% (10/12).
Notably, among 5 patients with TF expression ≥50% and intensity of 3+ treated at the 2 mg/kg dose, 4 achieved PR, and 1 achieved SD.
