Using mineralization-competent cells, this study examines how Sr²⁺ influences ECM mineral deposition by modulating ERK1/2–CREB pathways and regulating MV release, biophysical properties, and lipid composition. Advanced imaging, biophysical analyses, and state-of-the-art lipidomics reveal changes in vesicle mechanics and enrichment of ceramide and sphingomyelin, highlighting MVs as dynamic regulators of bone matrix formation.
