How important is ORF9b? More important, I think, than almost anyone has realized. ORF9b is often neglected—it is ignored on phylogenetic trees & on GISAID sequences. I see 9 lines of evidence indicating it's a major player. But they require some background. /1

So, what about non-canonical ORFs? We found peptides from #ORF9b and #ORF3c, revealing for the first time the contribution of internal ORFs to HLA-II peptides. As a side note, immunopeptidomics is a great tool to detect non-canonical proteins, often missed by proteome MS (ORF3c).

The overlapping gene #ORF9b was already present in all members of the species of SARS-CoV and SARS-CoV-2. See Figure 1 of our @elife paper ⬇️ ➡️https://t.co/5q1kDWghkp

A new report by @lucygthorne suggests the high transmissibility of SARS-CoV-2 #Alpha (B.1.1.7) involves the gene #ORF9b. 🔗https://t.co/e7qaqW35Pf ORF9b is an #OverlappingGene (#OLG)—a gene within a gene—shown below. As of now, no OLGs are annotated in the @NCBI reference genome.
