With roughly 20k human protein-coding genes and 100s of thousands of distinct proteins, understanding the intermediary—#RNAisoforms— is crucial for understanding human health and disease. Long-read sequencing (e.g., #ONT and #PacBio) makes their study possible, including the ~30k non-coding gene bodies.
Our work not only quantifies isoforms by tissue, but identifies genes that express many isoforms simultaneously, where 25 genes express ≥10 isoforms in cerebellum alone! We further validated the 447 new isoforms discovered in Aguzzoli-Heberle and Brandon et al. (including from new gene bodies).
RNA isoforms, though a known phenomenon, are incredibly understudied, and the #EbbertLab is setting out to help change that.
