We direct use the commercial solution for high-throughput imaging with add-on #SACDimaging executing. 10 min with 2 mm × 1.4 mm field-of-view for 128 nm overall resolution.

Then we applied #SACDimaging on a spinning-disk confocal (SDC) system (a Dragonfly system) for imaging deep 3D volumes (10 μm in depth with whole camera plane). XY/Z resolution: #SDC, 325 nm/655 nm; #SOFI, 296 nm/510 nm; #SACDimaging, 135 nm/334 nm.

Very happy to share our latest work, #SACDimaging, enhancing the fluorescence fluctuation detection sensitivity of SOFI by orders of magnitudes. It requires only 20 frames to achieve a >2-fold 3D resolution.
