New paper from Cornell @cornellvet by Mandy McGeachy on CD28 stimulation and effects on MEK1/2 and #JunB leads to overexpression of T-cell exhaustion markers #PD1 #TIM3 and increased IL-2 $CRVS data shows #ITK inhibition w/ #soquelitinib reduces #mTOR can lead to early stemlike Tpex generation, prevent T-cell exhaustion and lead to memory CD8+ T-cells. Also ITKi lowers MEK1/2 activity, reducing JunB, reduces IL-2 and reduces T-cell exhaustion markers (see paper below). While #OX40 and #Vav1 effects are briefly mentioned in paper. ITK inhibition has similar effects on pathway and its inclusion in paper could also further discussion on T-cell metabolism Akt-mTOR and T-cell exhaustion and memory cells. Paper even suggests: "Alternatively, one could test whether MEK1/2 inhibition along with anti-PD1 skews the effector to exhaustion ratio more favorably." This syngergistic effect is observed when ITK inhibition, which lowers ERK activity, is combined with immune checkpoint inhibitor like an anti-PD1.
