@plantmembranes @tophman13 @DrGriff34 I tried mRuby2 & mRuby3 in plants. mRuby2 was really bad, but mRuby3 was brighter and more stable than mCherry. But mRuby3 has disadvantages like blue-light activation, as mentioned by @DrGriff34. For me #mScarlet works the best: bright, quite stable & no photochromic behavior.

I'm not saying that lifeact doesn't have any effects - but if you want to study the effect of lifeact (rather than the FP) start out with a well-characterized monomeric fluorescent protein, e.g. mEGFP, mNeonGreen, #mTurquoise2, #mScarlet-I, mCherry
Fish people, I'm hearing good stories about #mScarlet(-I) and apparently #mTurquoise2 is also a well-behaving FP

Green: eGFP (with A206K) is good, as stable and good properties. If you want it very bright use YFPs (mCitrine, mVenus, or #sYFP2). all green/Yellow are not pH stable.
Alternative for pH stable is #mTurquoise2
@fpelisch Basics are important! Yes, #mTurquoise2 is possible with 405, but 440 is better (brighter, less autofluorescence).
488 also excte mTq2 a bit - we typically co-image with YFP or mNG using a 514 nm laser.
561 is great for #mScarlet-I or mCherry
