Paired with in vivo multiplexed screening, mBER forms @ManifoldBio's direct-to-vivo drug discovery platform, bridging the gap between AI-guided design and biological translation.
Excited to announce mBER, our fully open AI tool for de novo design of epitope-specific antibodies. To validate, we ran the largest de novo antibody experiment to date: >1M designs tested against 145 targets, measuring >100M interactions. We found specific binders for nearly half the targets, with up to 40% hit rates. Thread below:

This is absolutely brilliant work from our AI team in a rapidly advancing field. Equally impressive is the sheer scale and elegance of the experiment that validates the method (over 1M designs tested against 145 targets for a total of 100M data points!).
This is @geochurch-style ML^2 at it’s finest: Machine Learning with Multiplexed Libraries.
We built mBER to leverage the massive scale of our in vivo multiplexing technology for screening millions of designs directly in living systems.
Together, these form the foundation of Manifold’s direct-to-vivo drug discovery platform, bridging the gap between AI-guided design and biological translation.
In the limit, our goal is zero-shot drug design: From therapeutic profile to molecules that work in patients. To get there, we’re going to need new models trained on translationally-relevant data. We’re deploying mBER for brain delivery and other tissues. At the same time we’re generating semi-synthetic structure data backed by in vivo priors to advance this goal.
