Proud of the #PRG4 #lubricin manufacturing progress made working with @LubrisBio & @AvidBioservices the past 2+ years = a 500L GMP run. Eye drop bottles now filled as we push towards the clinic and 2 GRAMS (!) of #rhPRG4 leftover for future preclinical research the next 5+ years.

#PRG4 #lubricin AlphaLISA from @RevvityLifeSci! We developed it with @LubrisBio #rhPRG4, showed PRG4 down in Sjogrens tears, now great to have a kit made/optimized by Revvity CustomTeams. LLOQ ~5ng/ml, only 5ul of sample. Great for serum, tears, saliva - and we’ll see what else!

New lab paper, congrats to @ATang029 & thanks to coauthors! #PRG4 deficient mice have altered femur mechanical/structural/material properties, and more calvarial osteoclasts. AND, #rhPRG4 inhibits osteoclast-ogenesis & activity in vitro. In vivo is next?! https://t.co/Zw9ryCYXwj

Been working hard with team @LubrisBio that past 18 months on scaling up #biomanufacturing #rhPRG4. Lots of #WorldCourier shipments coast to coast, with some big boxes and hundreds of pounds of dry ice! Happy to be done for now, and can finally clear up some space in the lab 😊

#rhPRG4 is similar in size and charge to native #lubricin. ECF843 is smaller, more compact, less negatively charged, with altered disulfide bonding. Also less effective as a boundary lubricant and anti-adhesive, at inhibiting MMP9 activity, and slower to bind to collagen 1.(2/3)

New lab paper characterizing recombinant human #lubricin proteins! We demonstrate ECF843 has significantly altered size, charge, and diminished biological function compared to #rhPRG4. #Structure=#Function. We continue to study and develop rhPRG4 as a treatment for #dryeye.(1/3)
