Don’t reject a biomarker simply because it cannot be measured well in peripheral blood. Ask whether you are measuring the right analyte, in the right molecular form, in the right biological compartment.
A biomarker is not valuable simply because it is associated with disease or measurable at scale; it becomes translationally useful when the measured analyte is biologically interpretable, technically robust, and reproducible across datasets or orthogonal measurement layers.
For gene-silencing programs, “drug-targeted transcript” and “disease-relevant transcript” must be defined separately. They may overlap, but they are not necessarily identical.
Nonsense-mediated mRNA decay (NMD) guides brain development. When it fails, neurodevelopmental disorders follow. PTC position decides the outcome: haploinsufficiency vs. dominant-negative truncated protein. Key for variant interpretation & therapy. https://t.co/thv8RHH1XF
