Excited to share the final published version of my PhD work out today in @MolecularCell! See below for a thread of updates for the final version from the preprint! 1/ https://t.co/yAG3Nix5sI
Nice new preprint on oncogene alterations in lung adenocarcinoma that reinforces what we and others have seen with mEGFR and NKX2-1! https://t.co/lh6TxLPUro
ICYMI: since the preprint, we added extensive interrogation of NKX2-1 dosage to the final manuscript by both titrated lentiviral overexpression and knockdown, with distinct effects of dosage in the two contexts (OE: linear/thresholded, KD: mostly linear). https://t.co/yAG3Nix5sI
And if you're interested, check out our study on how NKX2-1 functions as a pioneer-like factor to drive lineage addiction through enhancer remodeling in lung adenocarcinoma! https://t.co/yAG3Nix5sI
It's a big week for NKX2-1!
Check out new studies on (1) how HNF4a modulates NKX2-1-mediated lineage specification (https://t.co/uJ6w6BsbAe) and (2) how NKX2-1 drives neuroendocrine differentiation in prostate cancer (https://t.co/oSEVBagqHM)
@jaybradner@Amgen Interesting to see the role of NKX2-1/TTF-1 here, we saw a role for it in EGFR persistence and speculated it might have a role for KRAS too!
Would be curious if NKX2-1 amplification status has any role here or if it’s just lineage state

@MolecularCell We've endeavored to make all the reagents and methods in this article as transparent and reproducible as possible. We deposited all plasmids (inc. pDONR cloning backbones and dTag OE destination vectors) on Addgene for easy use https://t.co/C9liTPasej 15/

@MolecularCell NKX2-1 is a complicated gene in cancer, with conflicting data for its role as a tumor suppressor and oncogene. Our data suggest NKX2-1 is a common oncogene and rare tumor suppressor in human LUAD, with it's TS role restricted to the rare IMA subtype 14/
