Interested in Ubiquitin, Parkinson’s disease therapy, mitochondria or DUB inhibitors? This preprint may be for you:
We show how the deubiquitinase USP30 can specifically be inhibited & introduce a framework for DUB ligandability.

Very excited to share that our manuscript on the "Discovery and mechanism of K63-linkage-directed deubiquitinase activity in USP53" was published today at Nature Chemical Biology @nchembio. Congratulations Kim, Kai, and all authors! https://t.co/YTV5td5koG
For all ubiquitin, deubiquitinase and chemical biology aficionados:
Here's a preprint from the lab introducing 2 new active DUBs in humans & a unique type of DUB activity one would miss with the widely used panels of free ubiquitin chains.

Excited to report our discovery of an important component of the oxidative stress response, the E3 ligase TRIP12. It acts as a chain elongation factor that amplifies CUL3-KEAP1 activity to drive NRF2 degradation as cells recover from stress.

Excited that our work on transcription-coupled DNA repair is now published @CellCellPress! We recapitulated TC-NER in Xenopus egg extract and identified STK19 as a core repair factor that couples stalled Pol II to TFIIH. 🧬🐸

Excited to share this work led by @CamillaElb
where we identify an entirely new role of LUBAC as a critical regulator of AMPK, the energetic stress response, and cellular metabolism. If you are interested in #ubiquitin and #metabolism, have a read: https://t.co/9pOwjckaVP

The study also establishes chimeric protein engineering to aid DUB crystallization – maybe a crystallization rescue strategy for other challenging enzymes, too?
We are excited to share this work & hope you will enjoy reading the preprint.
We look forward to comments!
This study is the result of persistent work and heroic efforts by Nafizul Kazi (on the far left in the picture). Chemistry by @NikolasKlink, cellular assays by @gallant_kai and Gian. Thank you for support by @mpimoph, @TU_Dortmund, @IMPRS_LM, @crc1430 .
