Kymera Therapeutics’ IRAK4 degrader, KT-474, was the first heterobifunctional degrader to show clinical activity in non-oncology indications, including atopic dermatitis and hidradenitis suppurativa, leading to its nomination as a 2023 Molecule of the Year.
Kymera’s partner, Sanofi, had initiated a number of Ph. 2 studies of KT-474 in HS and AD, restoring life to IRAK4 as an immunology target after Pfizer dropped its lead IRAK4 inhibitor, zimlovisertib (PF-06650833).
However on June 25, 2025, Sanofi announced that it plans to halt development of KT-474 and instead advance Kymera’s next-generation oral IRAK4 degrader development candidate, KT-485.
This updated article highlights the science behind the KT-474 program, including how it differentiates from small molecule inhibitors, its potential competitiveness with biologics and the first reported Cryo-EM ternary complex of a heterobifunctional degrader.
Despite its discontinuation, this 2023 Molecule of the Year Nominee will likely be considered a “landmark in drug discovery” for some time.
