I'm often asked how human genetics can be used to validate drug targets.
In our new @NatureCVR paper, we provide an end-to-end framework for IL-6 inhibition🧵
👉build a genetic proxy
👉demonstrate potential for cardiovascular benefit
👉assess expected & unexpected safety signals
"Don't bring me problems, bring me solutions" doesn't eliminate problems. It stops people from raising the hardest problems.
Leaders need to hear what's broken even if the fix is unclear.
A better option: “Bring me problems, as long as you’re willing to be part of the solution"
@tangming2005 The fact that you can plot that Venn is impressive Tommy. Although with such complex intersection, upset plot is obviously better for reading.
@tangming2005 Thanks Tommy, very useful thread as always. I would also add that mismatch gene id with gene symbol can happen with different annotation version as well. It might occur when you were handed an old dataset without proper documentation.
@MariosGeorgakis Thank you for an interesting article. It seems from panel D-E, youthful brain group has a lower risk of mortality -which is both surprising and positive.
@kjaganatha Thank you for sharing, Kishore. It looks very promising. One question, can we input a list of SNP (id or position) and check how they can influence gene expression?
@tangming2005 Very well said as always, Tommy. I would add 1 more thing: use seed number (set.seed) so you can reproduce result/plot as certain bioinformatics tools rely on randomness.
