Sample of clinical PROTACs in the degrader landscape
In advance of a bigger report on the space we're dropping tomorrow, some key takeaways from the space:
• Of the 53 clinical-stage oncology PROTACs with a https://t.co/ZKRHLP1b3Q reference (from 37 unique sponsors), 85% are in Phase 1 or 1/2. This is a classic wide + shallow space
• The latest stage assets target ER, AR, BTK and KRAS G12D. The first 3 build on established treatment markets, with an angle to pick up patients with resistance to existing drugs. The last one represents where the space wants to go: can degradation can produce a targeted therapy where none is approved today?
• Chinese-origin companies account for ~40% of this sample and 3/4 of the Ph2 programs. Meanwhile all 3 Ph3 programs have big Pharma involvement, as well as ~25% of programs overall
Look out for the full report tomorrow!
@JacobPlieth@raffcolo Mablink is just another story. The founders are from SKB Bio, which licensed the sac-TMT(TROP2) to Merck. Mablink team optimized the linker-payload and generated multiple assets. They have completed a lot of deals, such as Roche, ZLAB, Helius etc. So you can see many YL0014s
@JacobPlieth@raffcolo Warheads are all from Mabcare. CDH6 mab of CUSP06(AMT-707) is from Multitude and the ADC is also generated by Multitude. OnCusp licensed the global rights of AMT-707 except China right from Multitude in 2022. CUSP06 is second-in-class and potential best-in-class.
