I have only tried PEG as a molecular crowder for my experiments. Trying ionic solutes would definitely be something interesting to look at!
The Overhauser DNP data was taken at a magnetic field of 0.35 Tesla and electron Larmor frequency of 9.8 GHz.
Hydration water directly interacts with the protein interior or exterior and adopts different structures. The question we ask is whether the light-activated unfolding of AsLOV2 is driven by a thermodynamic action of water!

@asifequbal313@global_nmr When the pressure is from all sides though, like when we add just PEG (dark) at atmospheric pressure, it surrounds the protein from all sides and decreases the DEER distance a little compared to without PEG (dark) at atm.pressure.
@asifequbal313@global_nmr Also, the pressure is not high enough to completely denature the protein. We have tested reversibility when we de-pressurize it.
@asifequbal313@global_nmr The pressure is not coming from all sides. The setup is such that the pressure is exerted only from the top of the tube..thats why the deformation/compression of the helices leads to unfolding instead getting compressed into a ball.

The 17O NMR peaks were assigned as wrap and bound based on the DFT calculations of water clusters of model systems. @lokeswarnmr is working on the DFT part currently!
@asifequbal313@global_nmr The timescale for its light activation (from dark to light) is about 30 secs. And the dark state recovery takes around 1.5 min for the wild-type and 20 mins for the N414Q mutant.
@asifequbal313@global_nmr DEER data shows that light activation decreases the P(r) population around 2-3nm and increases it around 4-5nm. In the dark state, the average distance is 2.5nm. However, due to C-terminal disorder upon light activation, we can only estimate the lit state distance range as 4-5nm.
@asifequbal313@global_nmr additionally, we also see, light+pressure the effect is additive: more population in the longer distance. And also by adding PEG (molecular crowder) we are locally increasing more pressure, which also causes more population to go to the longer distance conformation.
