In the histograms above, it is pretty clear that the dimers are a majority of the hundreds of full-length reads, not a single artifact event, and the simplest explanation is the plasmid DNA itself is a dimer. You make a good point though!

Here is a nice example of the power of long-read full-plasmid sequencing such as the service offered by @SNPsaurus. We recently ordered a 4.7kbp plasmid from @Addgene. As part of our QC we sent it in for full-plasmid sequencing as part of a batch of plasmids... 1/n
@calinplesa@markwbudde We are doing 16 minimum right now, but since you are nearby just check in if you have smaller sets. We barcode and multiplex individual plasmids, can do complex pools if already barcoded on the plasmid.

@PgpMartin@AlbertVilella We do 384 plasmids at $15 a plasmid using @PacBio. Get HiFi full-length sequences that are polished to even higher quality with hundreds of other HiFi sequences from the same plasmid. Only downside is turnaround time (we do a run every other week) but working to get that better.
This article hits a pet peeve of mine. It says "RADSeq" in the title but it uses the ddRAD method. This is important because the study is a diverse genus of moth. Not a diverse species, but 43 samples and 32 species. Why does that matter? 1/n

@arunsethuraman For genomes under 500 Mb we think it makes sense to sequence the whole genome at 10X read depth. A 400 Mb beetle would be $87/sample for library prep, sequencing, and informatics. We would generate a draft reference from a few samples, align, call genotypes, return reads and vcf.
@PacBio keeps pushing the metrics. It has been great to work with a platform where we've seen multiple read-length doublings and 8-fold more reads in just the past few years.
