Proud to share that our latest work, „flexiAsCas12a and other Cas12a variants enhance the applicability of Cas12a nucleases in prime editing” is now available at Genome Biology.

Delighted to share our new method proPE (prime editing with prolonged editing window) is out in Nature Catalysis: https://t.co/JierTaoIvk
Kudos to first author Sarah Laura Krausz and all co-authors at HUN-REN TTK & HUN-REN SZBK! #CRISPR#primeediting
In our recent work published in @NatureComms we demonstrated a strategy to efficiently edit practically any target without off-targets using SpCas9 variants. The ‘CRISPRecise set' of variants will be available as a kit from @Addgene. @welkerlab @BiochemBrc https://t.co/a7KZ7GXWxE
@AltNHEJeh @biorxivpreprint Returning to this question, we checked and SuperFi-PE does not function with or without these mutations https://t.co/pK5liljSKW (Sup. Fig. 10). A possible explanation could be that the SuperFi H840A nickase (present in PE) shows very low activity (Fig. 4).
In our recent @biorxivpreprint we report that SuperFi-Cas9 exhibits extremely high fidelity but reduced activity in mammalian cells https://t.co/8aUbuL8Reo
In our new paper published in @NatureComms we characterize the on-target activity and off-target propensity of SuperFi-Cas9 in mammalian cells and also see strongly reduced activity but with high fidelity features. @BiochemBrc https://t.co/pK5liljSKW
In our recent @biorxivpreprint, using target-matched SpCas9 variants, we demonstrated efficient editing without off-targets on any SpCas9 target sequence, even on challenging targets that were attempted but failed in all previous studies. @BiochemBrc https://t.co/gGQ09UAGd7
@davidwtaylorjr Thank you David! Congratulations on your work! Your excellent structures added a few pieces to the incomplete puzzle of the SpCas9 mechanism.
@jeh_delivery @biorxivpreprint Thank you for your interest in our study! We are also puzzled by the lack of PE activity on targets where SuperFi nuclease is active and we can see your point, in fact we are already conducting this experiment alongside other approaches.
@JohnDoench@Fatwa_Adikusuma@jpkbravo@JohnDoench We checked and SuperFi is compatible with an additional 5' G at the end of the spacer (Fig. 1g-h,https://t.co/yLGnPOmAjc. We assume that it tolerates a 5' G because of the Y1013D mutation (same aa. mutated as in Blackjack variants- Kulcsar et al., Nat. Commun, 2020)
