Excited to share our systematic review and meta-analysis of standard- versus reduced-dose sotorasib in KRAS G12C-mutated NSCLC, now published in @JCOOP_ASCO
Reduced dosing did not appear to substantially compromise efficacy, supporting dose optimization efforts aligned with FDA Project Optimus.

Completely agree. Tolerability is more than adverse events. If reduced toxicity allows patients to stay on treatment longer without compromising target inhibition, it may become part of the efficacy pathway itself.
Grateful to @HannahRAbrams for highlighting this concept and to @NagashreeSeeth1 for her mentorship on the work behind these observations.
In this regard, drugs like Zofran, interventions like exercise, etc, are some of the most powerful cancer Rx we have. So kudos @Arthur_Aung on this study - higher upfront dose intensity is not always most effective if can’t consistently deliver it safely!
Excited to see this work presented at #ASCO26. Patients with pre-existing autoimmune disease remain underrepresented in immunotherapy trials, and real-world data are critical to informing treatment decisions. Looking forward to seeing the results and discussion.
