לראשונה פוענח המנגנון שמאפשר לסרטן השד ליצור גרורות במוח, אחת התופעות הקטלניות והקשות ביותר לטיפול בסרטן.

מחקר בינלאומי רחב היקף, בהובלת פרופ’ אורי בן דוד ופרופ’ רונית סצ’י פאינרו, יחד עם החוקרות ד”ר קתרין לאו וד”ר סבינה פוצי@sabina_pozzi ממעבדותיהם בפקולטה למדעי הרפואה והבריאות ע”ש גריי@TAUMedFaculty, חושף כיצד תאי סרטן השד מסתגלים לסביבת המוח, מנצלים את משאביה ומשגשגים בה.

ממצאי המחקר פורסמו בכתב העת היוקרתי Nature Genetics @NatureGenet

המחקר, שנערך בשיתוף פעולה עם עשרות חוקרים מארבע עשרה מעבדות בשש מדינות, זיהה שינוי כרומוזומלי מרכזי בתאי סרטן השד, פגיעה בגן p53 המכונה “שומר הגנום”, אשר מעלה משמעותית את הסיכון לגרורות במוח.

נמצא כי היעדרו של p53 תקין מאפשר לתאי הסרטן לשנות את חילוף החומרים שלהם, לייצר יותר חומצות שומן ולנצל חומרים המופרשים מתאי המוח לצורך הישרדות והתפשטות.

מעבר להבנת המנגנון, החוקרים בחנו גם תרופות ניסיוניות שמעכבות אנזים מפתח בתהליך, SCD1, וגילו שהן פוגעות באופן משמעותי בהתפתחות גרורות מוחיות, הן במודלים של עכברים והן בדגימות מנשים חולות סרטן שד.

לממצאים יש פוטנציאל ��שמעותי לפיתוח טיפולים חדשים ולרפואה מותאמת אישית. זיהוי מוקדם של הפגיעה ב p53 עשוי לאפשר התאמת טיפול ומעקב מדויק יותר, ולשפר את סיכויי ההחלמה של מטופלות בסיכון גבוה.

המחקר נתמך על ידי מענקי מחקר תחרותיים של הקרן הלאומית למדע ISF, הקרן לחקר הסרטן בישראל ICRF וקרן הבנק הספרדי Fundacion “La Caixa”. נוסף על כך, הוא מהווה חלק ממחקרים רחבים המתבצעים במעבדותיהם של פרופ’ רונית סצ’י פאינרו ופרופ’ אורי בן דוד, בתמיכת מענקי ERC מתקדמים, מענקי Proof of Concept וקרן קאהן.
