JBSNF-000088 is the one you actually want if the whole point is an oral NNMT inhibitor in a mouse lab-rat setup.
Funny, as I knew of this as a different name, but for the sake of this lol, happy to see it added as I had to rush to gather a bit more than I understood about it after I was given the heads up its hitting Elite!
Look at this as potentially being a much better version..... Though I have no lab-ratted it myself yet, and wont be for some time due to current plans.

5-Amino-1MQ gets the forum hype (which initially I was a fan of, and still kinda like the oral at high dose in a cut), but the published mouse work that made it famous was almost all subcutaneous (which is now hit or miss more than people think). When they finally measured oral in mice, it fell apart until doing was above cost worth for most
Both hit the same enzyme. NNMT methylates nicotinamide into 1-MNA and burns SAM doing it. Block it and you drop MNA, spare SAM, and in the fat-mouse papers you see less weight gain, smaller fat pads, better glucose tolerance, and lower triglycerides. That’s the whole “metabolic recomp” story people chase.
Where they split is the route.
JBSNF was built as a small nicotinamide analog and run oral from the start. In mice it hits about 40% oral bioavailability. 10 mg/kg oral gavage gives a Cmax around 3,500 ng/mL and peaks in half an hour. The diet-induced obese mouse paper dosed it 50 mg/kg twice a day by mouth for 4 weeks and got the body-weight drop, lower fed glucose, normalized oral glucose tolerance, less liver and plasma fat, and lower MNA in visceral fat.
Clean enough selectivity panel, negative Ames, soluble, and it actually absorbs.
5-Amino-1MQ is a charged quinolinium. Membrane-permeable enough for cells and for a needle, which is why the original Neelakantan obesity paper used 20 mg/kg subcutaneous three times a day and saw the fat-mass cut. Oral is a different animal. The 2024 mouse PK paper put oral bioavailability at about 3.5-10%. After 30 mg/kg by mouth the Cmax was only 14.5 ng/mL and it crawled in over 4 hours. High clearance in mouse hepatocytes, bad first-pass, poor enteric absorption. Rat data looks better (around 38% oral, longer half-life), but the efficacy literature everyone quotes is mouse, and in the mouse the oral number is garbage. So if you “just take it oral like the capsules,” you are mostly guessing against a species where the published oral exposure is tiny. Me personally, I still am/was a fan above 150mg, with proper deficit, AM initially. Even oral. I’m in minority there.
The catch on JBSNF is the half-life. About .4-.5 hours in mice, IV or oral. It gets in fast and leaves fast. That’s why the working protocol was twice daily, not once. 5-Amino hangs around longer when it actually gets into plasma (rat oral half-life around 7 hours), but that only matters if it absorbs. In the mouse oral setting it mostly doesn’t.
So the bro logic is simple. If the experiment is “oral NNMT inhibitor, mouse, look at weight and glucose,” JBSNF is the compound that was actually shown to do that by mouth, with real bioavailability and a published oral efficacy regimen. 5-Amino is the compound that works when you inject it and largely disappears when you feed it to a mouse. Short half-life means you dose JBSNF more often, but its likely 10x more impactful for the purpose would intend it for.
All in all, I'm seriously happy to see more orals popping up because that is the direction the market needs to go in the areas it can.
