⭐️Like in other hematologic diseases, our treatment targets are evolving:

From simply achieving a “safe platelet count”, we are moving towards 👇
➡️ patient-centred outcomes
➡️ better QoL
➡️ sustained remission
➡️ and, where possible, treatment-free remission and even cure.

Anita Kumar, @MSKCancerCenter, shares results from a phase II trial of zanubrutinib + obinutuzumab + venetoclax (BOVen) in older patients with previously untreated MCL (N = 50).
The MRD response-adapted BOVen triplet resulted in high rates of clinical and molecular response, and 2-year PFS and OS rates of 86% and 92%, respectively, in older patients with MCL. The regimen was well tolerated, with a safety profile consistent with previous reports.

María-Victoria Mateos shares extended follow-up data from the phase Ib RedirecTT-1 study of talquetamab + teclistamab at all doses (n = 94) and at the RP2D (n = 44) for the treatment of RRMM.

The ORR at all doses was 78% and 80% at the RP2D with 61% ≥CR. At the RP2D, 3-year PFS rate was 58% and OS was 74%. The safety profile was consistent with that of the known profiles for the individual agents.

CONGRESS | #ASH25 | PRESENTATION
Jennifer Brown shares results from an analysis of the ALPINE trial of zanubrutinib vs ibrutinib in patients with R/R CLL/SLL with ≥1 prior treatment, investigating symptom-based PFS as a clinically relevant and patient-centric endpoint (N = 608).

Analyses demonstrated significant associations between long-term patient-reported symptoms (fatigue, insomnia, and nausea/vomiting) and increased risk of disease progression. Risk of disease progression or death was ~30% lower with Zanubrutinib vs with ibrutinib across PRO symptom domains. Results highlight the potential for the use of patient-reported outcomes symptoms and symptom-based PFS as a composite endpoint in this patient population.
