We analyze the results of our protein design competition where 130 designers created binders for EGFR. With a 5x improvement in success rates and some designs outperforming clinical antibodies, we explore what worked, what didn't, and what this means for the future of protein engineering.

The long awaited results are here -- and they don’t disappoint!
📈 We doubled the number of proteins we tested in our lab from 200 to 400!
🧬 Out of those 400 proteins, 378 expressed (95% expression rate!)
🚀 Out of those 378 expressed proteins, 53 did successfully bind the target protein EGFR (that’s a 14% success rate, more than 5x of the success rate of the previous round just 2 months ago!)
💪 The best binders reached single-digit nanomolar affinities which is within the range of the commercially sold Cetuximab antibody by Merck
😎 30 out of 130 protein designers managed to design at least one binding protein

Incredibly excited to share new results from Nabla Bio where we show we can design antibodies de novo for use in therapeutic discovery.
We introduce JAM, an AI system we’ve developed to design de novo antibodies with good affinities, early stage developability, and function. We’re seeing success across a range of soluble and hard-to-drug membrane proteins including a Claudin and a GPCR. We’ve extensively tested these designs in our wet lab and included detailed data and controls, providing the first clear demonstration of how de novo design could expand the scope and efficiency of therapeutic antibody discovery. 🧵
