We show that multiple defects caused by mutations of a conserved subdomain of the catalytic core, including impaired growth and DNA synthesis and genome instability, were rescued by exonuclease inactivation. 2/3

We show that ORC hypersumoylation preferentially reduces the function of a subset of early origins, while Orc2 hyposumoylation has the opposite effect. Mechanistically, ORC hypersumoylation reduces MCM loading in vitro and diminishes MCM chromatin association in vivo. 2/3
