@andrewcaravello I think the key is how strong and durable the abscopal effects from DCVax vs other cheaper competitors such as:
(1) intratumor injections of molecule tigilanol tiglate (Stelfonta), phase 2a clinical trial (NCT05755113)
(2) Intratumoral oncolytic virus (OV) therapies

For other cancer types patients have multiple solid mets in the body, Seems like DCVax mainly rely on intratumor injections to treat accessible tumors. Does DCVax injection trigger abscopal effects to kill other remote mets?

If DCVax is so powerful, why it only extend patients life 3 months ? What limit its effectiveness?

Efficacy depend heavily on the underlying cause of immune deficiency (age, radiation, infection, lymphopenia source, immunosuppressive therapies, etc.). So IL-7 likely won’t “solve all immunodeficiency” by itself.
@andrewcaravello Because IL-7 broadly stimulates T cells (naïve, memory, effector), there’s a concern that it might also expand unwanted or autoreactive clones (especially in autoimmune-prone individuals). Indeed, IL-7/IL-7R-pathways are implicated in autoimmunity.

IL-7 therapy has Transient effects: In some models (e.g., aged primates), IL-7 therapy increased naïve T cell counts only transiently — often via expansion of existing peripheral T cells rather than renewed thymic output — limiting long-term restoration.
@andrewcaravello Such lymphopenia most likely contributes to aggressive growth of other Mets in cancer patients body after bone marrow damage from radiation ( some CAR-T immunotherapy also cause lymphopenia) . Any IL-7 based solution for restore immune function, not just ALC number ?
@andrewcaravello Actually most chemo did not cause persistent low ALC , just cause temporary low neutrophil count. what really cause lymphopenia is radiation therapy when the radiation beam go thru bone and bone marrow which is very sensitive to very low radiation dose. The ALC is hard to recover
@andrewcaravello@doc_brown2@NewsNation@drpatrick Cancer patients have no time to wait, both DCVax and Anktiva should be approved by FDA asap for all cancer types. The enemy is FDA and the evil force behind FDA. Cancer needs multiple solutions to work together, mono-therapy never works !
@andrewcaravello Our so called scientists or science may only know less than 10% how our body immune system works. So as long as the drug is safe and FDA has no reason to restrict dying cancer patients to try it, and let the data and common sense talk.
