Despite distinct genetic and transcriptional programs, tRCC and ccRCC share a VCAM1⁺ proximal tubule origin. Some tRCCs, however, may arise from other kidney lineages - plasticity that could explain its varied histology and presence of TFE3 fusions in ASPS and PEComa.

We generated the first ever, murine transgenic kidney tumor models of SFPQ-TFE3 - the most common MiT/TFE gene fusion implicated in both, tRCC and PEComas (Perivascular Epitheloid neoplasms).
We describe a novel role for this fusion in regulating #mTORC1-driven lineage plasticity, with transcriptional downregulation of critical nephric lineage transcription factors like PAX8, that perfectly recapitulate human renal PEComas.
Additionally, we nominate the lysosomal V-ATPase complex as a central regulator of mTORC1 activity in tRCC, and a target-rich environment for therapeutic intervention in this deadly disease.

Life can be incredibly cruel. 14, what a beautiful age. Austen from Missouri was diagnosed with TFE3 rearranged PEComa overlapping with tRCC only 5 months ago. She passed away this morning. This hurts like hell.
Much respect to @srviswanathan recipient of Kidney Cancer Innovation/Discovery Grants! Rare subtype like tRCC often received less funding and attention. It takes dedication, creativity, and compassion to tackle diseases that are poorly understood and harder to treat. #KCRS25
