William Wierda, @MDAndersonNews, presents post hoc exploratory analysis results of the phase I/II TRANSCEND CLL 004 study evaluating characteristics associated with response to liso-cel in pts with R/R CLL/SLL (n=87). OR and CR rates did not correlate with age and high-risk features at screening including unmutated IGHV, TP53m, 1del(7p), or complex karyotype. Patients receiving ≤3 prior treatments had numerically higher OR rate (54.5%) than those receiving >3 prior treatments (44.6%).

William Wierda, @MDAndersonNews, discusses whether continuous therapy is the best option for high-risk pts with CLL. In pts with del(17p)/TP53 mutations, second generation covalent BTK inhibitors (such as ibrutinib) show median PFS of 6+ years, however, are associated with continuous risk of AEs or toxicities. Other options include non-covalent BTK inhibitors & BCL2 inhibitors.
