The review is the product of truly great, inspirational and collaborative endeavor with our 2 PhD students (Flore Sneyers & Femke Speelman-Rooms) and our ChemBio partner @VerhelstLab and Calcium partner @MartinBootman! 🙏for your help, input & feedback throughout the work!
Science requires proper controls. BAPTA evoked MCL-1 downregulation, so "Ca2+ drives MCL-1 abundance"? It turned out that TF-BAPTA (Kd 65 µM, far off 100 nM basal [Ca2+]) too evoked MCL-1 decline! So, Ca2+ chelation was not the cause. Our journey led to direct PFKFB3 inhibition!

It wasn't easy and risky working on an "off-target". But, we found it worthwhile to properly interpret findings as BAPTA is used in >1000s studies but also learning new things such as PFKFB3 linking to MCL-1.
Happy to share @cddpress paper by Flore Sneyers identifying PFKFB3 enzyme as direct target of intracellular Ca2+ chelator, thereby accounting for Ca2+-independent inhibition of mTORC1, MCL-1 downregulation and cell death in MCL-1-addicted cells!

@danmjohnson@JMCCardiology@ChrisToepfer Interesting study! Unfortunately, there are wider cellular issues with calcium indicators and BAPTA too. https://t.co/no9MBdB6PA doi: https://t.co/lVsSn7hWqM
@jaketapper Best wishes to you and your family Jake. With your daughter’s recent traumatic health issue, and now losing Tom, you’ve been through a lot. I can empathise with your situation and loss. Sincere sympathy to you all.
A special word to … @MartinBootman. Thanks AV-man! 🤣 🙏 @MaleneBrohus and @pchoern for many small & big things and your continuing enthusiasm & support with @EuroCalcium Board! 🙏 @T_Luyten for all your help throughout! Definition of friends: they help out without asking! 🙏
Jeez. In the first 30 minutes of tonight’s Tory leadership debate Rishi Sunak & Liz Truss managed to completely eviscerate the whole Tory Government
