In the Triple Threat framework, the 2026 eBioMedicine study showing significantly reduced striatal VMAT2 binding in patients with Spike Pathology provides direct imaging evidence of dopaminergic terminal injury that aligns precisely with the convergent effects of the three circuits on vulnerable monoaminergic neurons.

The hibernation-like torpor circuit suppresses mitochondrial function and ATP production in striatal neurons, impairing the energy-intensive process of packaging dopamine into vesicles via VMAT2. This leads to reduced vesicular storage capacity, lower synaptic dopamine release, and the observed PET signal loss, which correlates with apathy in the ventral striatum, motor slowing in the dorsal putamen, and memory deficits in the caudate.

The immune tolerance circuit prevents effective immune clearance of damaged dopaminergic neurons and protein aggregates, allowing senescent cells and low-grade neuroinflammation to persist. This creates a chronic, non-resolving state where partial dopaminergic dysfunction becomes self-sustaining rather than transient.

Together, the three circuits produce a measurable reduction in presynaptic dopaminergic integrity without requiring outright neuronal death, explaining the specific neuropsychiatric profile of apathy, bradykinesia, and cognitive slowing.

VMAT2 is also central to the controversial “god gene” hypothesis proposed by Dean Hamer in 2004. Hamer suggested that certain variants of the VMAT2 gene (SLC18A2) correlate with higher scores on Cloninger’s self-transcendence scale, which measures tendencies toward absorption, connectedness to a larger universe, and openness to mystical experiences. The proposed mechanism involves altered monoamine signaling (dopamine, serotonin, norepinephrine) that influences higher-order perception and emotional tone.

In this framework, Spike Protein–induced reduction in striatal VMAT2 function would not only impair motivation and movement but could theoretically blunt or distort the monoaminergic tone that supports a sense of self-transcendence or spiritual connectedness. Individuals with persistent Spike Protein pathology might therefore experience a dampened capacity for reward, meaning, or spiritual experience alongside the more obvious fatigue and cognitive symptoms, a higher-order manifestation of the same dopaminergic terminal injury documented in the PET study.

This integrated view positions the observed VMAT2 reduction as one measurable downstream consequence of the Triple Threat state acting on the brain’s reward and motivational circuitry.
