BREAKING NEWS
The 2025 #NobelPrize in Physiology or Medicine has been awarded to Mary E. Brunkow, Fred Ramsdell and Shimon Sakaguchi โfor their discoveries concerning peripheral immune tolerance.โ
๐๐ผ๐๐ผLetโs be mindful of how we define macrophages in tumors. They are not M1 or M2. In order to move the field forward we need to use better description of this highly heterogeneous population of cells.
PSA: Stop calling macrophages in tissue M1 or M2. These are not states that exist in biology. The only use of M1 is a macrophage cultured ex vivo with LPS and IFNg; and for M2: IL-4/13/10. Macrophages in tissues are highly complex and diverse and do not resemble either of the aforementioned M1/M2 states. M1/M2 language causes confusion and sets the field back. Refer to your macrophages by the molecules they express and the cytokines they make.
๐ง ๐ฅ Can macrophages revolutionize cancer immunotherapy?
Macrophages naturally infiltrate tumors, reshape the TME, phagocytose cancer cells ๐ฆ , and activate T cells โ๏ธโmaking them powerful tools for next-gen therapy.
๐ Enter CAR-Ms: Chimeric Antigen Receptorโengineered Macrophages.
๐ฌ In early trials and preclinical studies, CAR-Ms:
โ Kill tumors via phagocytosis
โ Modulate immunosuppressive TMEs
โ Induce endogenous anti-tumor T cell responses
โ Show low CRS risk vs CAR-T cells
โ Infiltrate solid tumors more efficiently than CAR-T/NK cells
๐งช Key strategies boosting efficacy:
Anti-CD47 ("donโt eat me" blockade)
Anti-PD-1 combination
Proinflammatory signaling (e.g. IL-12, IFN-ฮณ)
In vivo delivery via LNPs or hydrogel platforms
๐ก CAR-Ms may solve CAR-Tโs biggest limitations:
โ ๐ง Toxicity
โ ๐ฐ Cost
โ ๐ Solid tumor resistance
๐ฏ Future focus:
Safer vectors (CRISPR > lentivirus)
Off-the-shelf or in vivo platforms
Targeting oncogenic drivers, not just surface antigens
