I’ve admired @AnnaEnim since her original GoT - so much so that I flew from AUS to Cornell just to meet her in person (she’s learning this now!). Instant connection. Many coffees, visits, and brainstorming years later…

-What’s new
We developed Genotyping of Transcriptomes for multiple targets and sample types (GoT-Multi) - a next-gen single-cell multi-omics platform that:
•Links somatic genotypes to whole-transcriptome states in thousands of cells
•Works beautifully on FFPE tissues (hello, clinical archives!)
•Uses probe-based ligation + ML-optimized genotyping
In short - high-throughput, scalable, and accessible single-cell genotype–phenotype mapping.
-What we found
Applying GoT-Multi to Richter transformation (CLL → aggressive lymphoma):
•Mapped 27 mutations across thousands of single cells
•Reconstructed clonal architectures and their transcriptional programs
•Found that distinct genotypes - including therapy-resistant subclones - converge on an inflammatory state
•Discovered hints of subclonal cooperation, where small clones may actually support the fitness of others
Different genotypes. Shared transcriptional destiny. That’s how cancer learns to resist.
-Why it matters
GoT-Multi finally bridges the gap between clonal architecture and cell-state heterogeneity - directly, scalably, and even from FFPE samples.
It opens the door to retrospective clonal tracking, precision biomarker discovery, and spatial-genotypic integration.
Proud, grateful, and honestly blown away by what this collaboration achieved. None of this wouldn’t have happened with the amazing talents of Minwoo and Mirca…two incredible talented next generation scientists!
